Tango drug combination shows 92% response rate in pancreatic cancer trial

Tango Therapeutics Inc. (NASDAQ: TNGX) reported that a combination of its experimental drug vopimetostat with Revolution Medicines' daraxonrasib achieved a 92% objective response rate in patients with previously treated pancreatic cancer in an ongoing Phase 1/2 trial.
The combination demonstrated a 90% six-month progression-free survival rate among 12 evaluable patients with pancreatic ductal adenocarcinoma who had MTAP deletions and RAS mutations, according to data from the clinical trial as of May 28, 2026.
All patients in the study had advanced disease, with 70% having liver metastases, and more than half received the combination as third-line treatment. The treatment showed a 100% disease control rate in the pancreatic cancer patients.
In a separate arm of the study, vopimetostat combined with Revolution Medicines' zoldonrasib showed a 52% objective response rate and 74% six-month progression-free survival rate among 27 evaluable pancreatic cancer patients.
The combinations were generally well-tolerated, with most adverse events being Grade 1 or 2 in severity. The most common treatment-related side effects for the vopimetostat-daraxonrasib combination were rash, stomatitis and diarrhea, while nausea and vomiting were most common with the zoldonrasib combination.
Based on the results, Tango plans to advance the vopimetostat-daraxonrasib combination into Phase 3 development for first-line treatment of MTAP-deleted pancreatic cancer. The company expects to finalize the Phase 3 trial design in the second half of 2026.
Vopimetostat is an experimental PRMT5 inhibitor designed to work selectively in cancer cells with MTAP deletions, which occur in approximately 40% of pancreatic cancers and 15% of lung cancers.
