The analyst comments "What’s new? We are initiating coverage of Immunome with a Buy rating and a $40 price target, reflecting our conviction that the company is building a portfolio of best-in-class, pipeline-in-a-product oncology assets with meaningful differentiation across both validated and underpenetrated solid tumor and hematologic indications. In this report, we evaluate the core drivers of value through a scientific and commercially grounded lens, focusing on Immunome’s mechanistic advantages, discovery engine productivity, and ability to translate platform innovation into durable clinical and revenue growth. We view lead asset varegacestat as a best-in-class gamma-secretase inhibitor for desmoid tumors, demonstrating superior median tumor volume regression, Progression Free Survival (PFS), and Overall Response Rate (ORR) versus the only approved therapy, Ogsiveo, and we believe its differentiated efficacy and tolerability profile position it to become the category leader with peak sales that could exceed $1B by 2034 (NDA submission 2Q26). Beyond varegacestat, Immunome is assembling a next-generation ADC franchise led by the ROR1-targeted IM-1021, currently in Phase 1/2 studies across relapsed/refractory B-cell lymphomas and advanced solid tumors with topline data expected in 2026. The company’s ADC approach pairs underappreciated targets with optimized linker and payload chemistry enabled by a high-throughput discovery engine, highlighted by the proprietary TOP1 inhibitor HC74, designed to overcome chemoresistance while preserving a robust bystander effect. With multiple additional ADC INDs planned through 2026 and longer-term expansion via IM-3050, a FAP-targeted radioligand therapy for FAP-expressing solid tumors, we see Immunome’s portfolio as a coordinated set of value drivers, anchored by near-term execution from varegacestat and sustained medium- to long-term growth from the ADC and radioligand platforms."
