Zymeworks outlines 2026 strategy with potential $440M milestones

Zymeworks Inc. (NASDAQ: ZYME) announced its strategic priorities for 2026, highlighting potential milestone payments of up to $440 million related to regulatory approvals of its cancer drug Ziihera in gastroesophageal adenocarcinoma across multiple regions.

The biotechnology company reported positive Phase 3 results for Ziihera (zanidatamab-hrii) in first-line HER2-positive gastroesophageal adenocarcinoma, presented at the ASCO Gastrointestinal Cancers Symposium. The trial showed statistically significant improvement in progression-free survival compared to standard treatment with trastuzumab and chemotherapy.

Zymeworks said it could receive milestone payments totaling $440 million for potential regulatory approvals: $250 million from the United States, $100 million from Europe, $75 million from Japan, and $15 million from China. Partner Jazz Pharmaceuticals plans to submit a supplemental Biologics License Application in the first half of 2026 for zanidatamab in the U.S.

The company reported cash, cash equivalents, and marketable securities of approximately $270.6 million as of December 31, 2025. Combined with anticipated regulatory milestone payments, Zymeworks expects sufficient cash runway beyond 2028.

In November 2025, the board authorized a $125 million share repurchase program. The company has utilized approximately $19 million to acquire 727,271 shares at an average price of $26.07.

Zymeworks earned $69.6 million in milestone payments in 2025 from partners including Bristol Myers Squibb, GlaxoSmithKline, Johnson & Johnson, Daiichi Sankyo, and BeOne Medicines from zanidatamab and legacy platform collaboration agreements.

The company plans to continue Phase 1 clinical studies for drug candidates ZW191 and ZW251 during 2026, while advancing other research efforts primarily through partnerships. Zymeworks expects to file investigational new drug applications for multispecific programs ZW209 and ZW1528 in 2026.
