Akari Therapeutics outlines 2025 progress in shareholder letter

Akari Therapeutics Plc (NASDAQ: AKTX) released a shareholder letter detailing the company's 2025 developments in its antibody drug conjugate programs. The oncology biotechnology company reported progress on its lead program AKTX-101 and manufacturing partnerships.

The company initiated a manufacturing partnership with WuXi XDC to produce clinical-grade material for AKTX-101, which targets the Trop2 receptor using Akari's proprietary PH1 payload. The partnership aims to support planned Phase 1 clinical trials scheduled for late 2026 or early 2027, subject to regulatory clearance.

Akari reported preclinical data showing AKTX-101 demonstrated activity in pancreatic cancer models driven by KRAS mutations and against AR-V7 in prostate cancer models. The company previously reported activity in urothelial and gastric cancer preclinical models.

The company filed three new patent applications related to its ADC payload technology in the last two quarters of 2025. Akari also presented data at the Society for Immunotherapy Cancer conference, which was noted among the top 150 submissions.

AKTX-101 uses a spliceosome-modulating mechanism through its PH1 payload, which differs from traditional ADC payloads that typically use tubulin inhibitors or DNA damaging agents. The company established a scientific advisory board to guide development of its PH1 payload platform.

Akari outlined planned 2026 milestones including regulatory interactions with the FDA, presentation of comparative data against other Trop2 ADCs, completion of chemistry, manufacturing and controls work, and potential initiation of Phase 1 trials.
