H.C. Wainwright analyst Joseph Pantginis reiterated a Buy rating and $4.00 price target on Lexicon Pharmaceuticals (NASDAQ: LXRX).

The analyst commented: "At the International Congress on Neuropathic Pain (NeuPSIG) 2025, Lexicon presented new preclinical data demonstrating pilavapadin-mediated neuropathic pain (NP) reductions in animal models of chemotherapy-induced NP (CINP) and multiple sclerosis (MS). We believe these early preclinical observations: 1) highlight the relevance of the serine/threonine kinase, adaptor-associated kinase 1 (AAK1), as a regulator of pain signaling; 2) validate pilavapadin (AAK1 inhibitor; formerly known as LX9211) as a non-opioid therapeutic candidate for diverse chronic NP conditions; and 3) support further clinical evaluation of pilavapadin in CINP and MS, along with diabetic peripheral neuropathic pain (DPNP; see favorable topline results from the Phase 2b PROGRESS dose-optimization trial in our prior note: PROGRESS Identifies Optimal Dose with Meaningful DPNP Reductions; Pilavapadin (LX9211) Phase 3 Initiation in 2025). We foresee pilavapadin aligning with the recently published FDA’s draft guidance, entitled “Development of Non-Opioid Analgesics for Chronic Pain” (publication date, September 10, 2025), in which the agency urges the acceleration of safe and effective non-opioid treatments and the reduction of prescription-related opioid misuse, by developing novel therapies that cover multiple chronic pain conditions rather than individual indications. CINP is a common, debilitating, and often persistent side effect associated with chemotherapeutic agents such as paclitaxel, oxaliplatin, and bortezomib. Underlying pathophysiology involves mitochondrial dysfunction, axonal degeneration, and neuroinflammation. It affects up to 68% of patients during treatment, and can severely impair QoL (Seretny M., et al. Pain. 2014). Similarly, 50–75% MS patients experience NP due to demyelinating lesions, characterized by burning sensations, sharp shooting pain, tingling, numbness, and hypersensitivity to stimuli (Ouyang, H., et al. Front Immunol. 2024). Reiterate Buy rating and $4 PT."
