Cantor Fitzgerald analyst Dr Charles Duncan reiterated an Overweight rating and $43.00 price target on Alkermes (NASDAQ: ALKS).
The analyst commented, “We are providing a brief preview for the P2 Vibrance-1 (NT1, early-3Q25) and Vibrance-2 (NT2, data Fall’25) trial readouts. The data will be pivotal in de-risking ALKS 2680’s profile as a highly selective, QD oral orexin 2 agonist (we say “OX2Rag”) with potential best-in-class differentiation. The studies may validate ‘2680’s therapeutic index, leveraging its high selectivity/potency and safety/tolerability profile, which could address unmet needs in both NT1 (orexin-deficient) and NT2 (non-orexin pathophysiology) populations. While TAK’s (NC) TAK-861 (P3 program ongoing in NT1) leads the class, ‘2680’s broader development in NT1, NT2, and IH and supportive P1b data (though in small sample sizes) suggest competitive positioning. As such, we would like to see a similar improvement in Measurement of Wakefulness Test (MWT) from baseline seen from the P1b demonstrated in the P2 trials for NT1 and NT2, respectively. Specifically, this is a mean sleep latency of ~30 min in Vibrance-1 (with the 8mg dose) and >25 min in Vibrance-2 (18mg). Safety remains a focus given common AEs of visual disturbances and on the more extreme end, TAK-994’s hepatotoxicity, but ‘2680’s P1b showed no severe AEs (mostly mild insomnia, pollakiuria), likely indicating tolerability in the polypharmacy-heavy pt pop.”
